Detecting beta-amyloid glycation by intrinsic fluorescence - Understanding the link between diabetes and Alzheimer's disease

چکیده
We monitor early stages of beta-amyloid (A? 1–40 ) aggregation, one the key processes leading to Alzheimer's disease (AD), in presence high glucose concentrations by measuring A? 1– 40 intrinsic fluorescence. The multiple peaks and their shifts observed time-resolved emission spectra (TRES) reveal impact glycation on oligomerisation. results show that formation advanced end products (AGEs) alters aggregation pathway. These changes are highly relevant our understanding pathophysiology AD implication AGE diabetes these pathways.

متن کاملAlzheimers Disease: A Novel Hypothesis for the Development and the Subsequent Role of Beta Amyloid
Spirochetes, biofilms, and innate immune system activity have all been recently found in the brains of Alzheimer’s disease patients. The mechanism and actions of those factors in producing the disease were discussed in those studies. However, neither the production nor the role of beta amyloid was included in the discussion of those studies. In this commentary, we hypothesize how the developmen...
متن کاملGlycation of the amyloid beta-protein by a nicotine metabolite: a fortuitous chemical dynamic between smoking and Alzheimer's disease.
The origin of Alzheimer's disease (AD) has been subjected to an intense amount of examination; however, a clear conclusion as to the nature of this crippling disease has yet to be identified. What is readily accepted is that a definitive marker of this disease is the aggregation of the amyloid beta-peptide (A beta) into neuritic plaques. The recent observation that nicotine exposure leads to de...
متن کاملInsulin Resistance as a Link between Amyloid-Beta and Tau Pathologies in Alzheimer’s Disease
Current hypotheses and theories regarding the pathogenesis of Alzheimer's disease (AD) heavily implicate brain insulin resistance (IR) as a key factor. Despite the many well-validated metrics for systemic IR, the absence of biomarkers for brain-specific IR represents a translational gap that has hindered its study in living humans. In our lab, we have been working to develop biomarkers that ref...
متن کاملP135: The Role of Amyloid Beta-Peptides and Tau Protein in Alzheimer\'s Disease
Alzheimer's desease is the most common age-related neurodegenerative disorder, and cognitive problems such as defects in learning and memory are of its symptoms. Among the factors involved in the pathogenesis of the disease are biochemical disorders in protein production, oxidative stress, decreased acetylcholine secretion and inflammation of the brain tissue. Extra-neuronal accumulation ...
